Preclinical Modeling of Clinically Relevant Radiation Therapy for Treatment of High-Risk Medulloblastoma
- Abstract:
- Group 3 medulloblastoma (MBGrp3) with MYC overexpression and Sonic hedgehog medulloblastoma (MBSHH) with MYCN amplification and TP53 loss of function represent high risk MB with particularly poor prognosis. Despite aggressive multimodality therapy with high dose craniospinal irradiation (CSI) and adjuvant chemotherapy, a significant number of high risk patients fail primary therapy. The objective of this study was to initiate preclinical trials designed to more faithfully recapitulate aspects of the human disease and therapeutic interventions for high-risk MBs in order to systematically test novel combination therapies that may be translated to the clinic.
