Figure 2 from Imaging Glioblastoma Response to Radiotherapy Using 2H Magnetic Resonance Spectroscopy Measurements of Fumarate Metabolism

Abstract:
<p><sup>2</sup>H MR spectroscopy measurements of labeled fumarate, malate, and water concentrations in A11 (<b>A–E</b>), S2 (<b>F–J</b>), and U87 (<b>K–O</b>) tumors. Tumor spectra were acquired before and 48 hours after the last chemoradiotherapy treatment (temozolomide 100 mg/kg, 20 Gy in total, 5 Gy per fraction). Sum of 12 <sup>2</sup>H spectra recorded over 60 minutes from A11 (<b>B</b> and <b>D</b>), S2 (<b>G</b> and <b>I</b>), and U87 (<b>L</b> and <b>N</b>) tumors. Injection of [2,3-<sup>2</sup>H<sub>2</sub>]fumarate (1 g/kg) started 5 minutes after the start of acquisition of the first spectrum. The peaks were fitted individually using prior knowledge. [2,3-<sup>2</sup>H<sub>2</sub>]malate/[2,3-<sup>2</sup>H<sub>2</sub>]fumarate ratios before and 48 hours after the last chemoradiotherapy treatment of A11 (<i>n</i> = 3; *, <i>P</i> = 0.0439, paired two-tailed <i>t</i> test; <b>E</b>), S2 (<i>n</i> = 3; ***, <i>P</i> = 0.0006, paired two-tailed <i>t</i> test; <b>J</b>), and U87 (<i>n</i> = 3; **, <i>P</i> = 0.0061, paired two-tailed <i>t</i> test; <b>O</b>). Data are presented as mean ± SD.</p>
