- The ability to express proteins in heterologous systems has been crucial to the biochemical analysis of many cell cycle regulators. Until recently, bacterial expression systems have reigned supreme due to their ease of use and their ability to produce vast quantities of a desired protein. However, many proteins are recovered from bacteria in an inactive form due either to improper folding or to lack of critical covalent modifications (e.g., phosphorylation or glycosylation).
